本課題組致力于設計開發具有特定功能的人造蛋白材料,以滿足當前資源、能源、環境等可持續發展策略對新材料的迫切需求。本實驗室通過將生物物理、生物工程以及醫學等領域進行交叉融合,建立了以蛋白功能為導向、以生物合成和多尺度模擬為基礎、以高通量制備分析和功能測試為驗證和優化手段的人造蛋白功能材料創制體系。主要研究方向為(1)蛋白質序列-結構-功能的關系以及蛋白質材料對人體組織/細胞生長分化的影響;(2)智能蛋白質材料設計、高通量篩選、蛋白質自組裝理論研究和蛋白質材料物理化學改性;(3)蛋白質基柔性電子設備研發等應用方面的研究工作。

Dr. Huang’s lab focuses on predictive design, synthesis and functionalization of protein-based biomaterials for regenerative medicine and sustainable bio-electronics. Current research projects include: (1) injectable hydrogel and stem cell engineering, (2) stimuli-responsive biomaterial design, and (3) flexible electronics. Postdoc positions are available for highly motivated candidates with expertise in synthetic biology, material science and stem cell engineering.

陳迪博士的實驗室研究人類胚胎發育和胚胎干細胞分化的調控。目前,陳迪實驗室招聘數位博后,主要研究課題包括:

Dr. Chen’s lab focuses on the regulation of human embryogenesis and embryonic stem cell differentiation. Postdoc positions are available in Chen lab for the following projects

(1) The regulation of human embryonic stem cell differentiation at epigenetic and post-transcriptional levels.

(2) The function of RNA-binding protein during human primordial germ cell specification and differentiation.

(3) The new functions of cell fate determinant transcription factors during embryonic stem cell differentiation.

研究領域Research: The Leptihn Group offers two Postdoctoral Fellowships; one project focusses on the investigation of membrane proteins controlling cellular signalling pathways employing microfluidics and single molecule methods (fluorescence, electrophysiology) together with the University of Edinburgh and the ZJUI (Zhejiang University-University of Illinois Urbana Champagne). The second project involves viruses of microbes (bacteriophages) with aspects on both, function and structure of phage proteins, as well as phage therapy. Experience in imaging and protein biochemistry or microbiology are desired.

本課題組將從細胞器交流角度解析細胞-外界環境的相互作用,即外界環境變化(特指外源的能量供給變化)對細胞器互作結構特質(property)/穩態(homeostasis)的影響。直接研究方向主要集中在兩個方面:1)結合領域內已知和本人的前期數據,對細胞器互作網絡改變細胞器膜脂組成和蛋白成分進行深入的機制研究。2)利用新的生物標記或技術從亞細胞器水平上發現細胞器互作網絡對單個細胞器的影響,進一步拓展我們對細胞器互作網絡的功能認知。

The main focus of our lab is to elucidate the molecular pathways and cellular interactions that mediate and regulate thymocyte development and T cell-mediated immune responses by using molecular biology and genetic approaches. 郭博士從事干細胞和癌癥的研究,精于人類和小鼠遺傳學,分子生物學,細胞生物學,生物醫學,及基因組學的方法。他在造血發育、白血病干細胞以及腫瘤耐藥調控方面等有重要建樹。他建立了第一個由PTEN缺失導致T細胞急性白血病的小鼠模型,促成臨床發現只帶有PTEN突變而沒有NOTCH1突變的嬰幼兒T細胞白血病病人。他首次分離了T細胞急性白血病干細胞,揭示出了多因子突變導致白血病干細胞形成的過程,并發現了mTOR抑制劑的雙重抗癌機制。近來,郭博士課題組從臨床肺腫瘤中分離出腫瘤干細胞,并發現NOTCH1信號通路調控肺腫瘤干細胞的自我更新和鉑類耐藥。

Dr. Wei Guo laboratory uses basic biological approaches to understand biomedical challenges and translates its understanding to clinical practice. There are two major research directions: (1) stress response and ex vivo expansion of hematopoietic stem cells; and (2) regulation and targeted inhibition of cancer resistance.

研究方向:課題組運用分子生物學、細胞生物學、生物信息學、3D類器官培養和轉基因小鼠模型研究干細胞命運控制,衰老及相關疾病的靶向治療。

正在開展的課題:

1)腸干細胞穩態平衡、惡性轉化及腸道疾病的早期診療。運用生物信息學分析、小鼠模型和腸道類器官,我們將深入研究大腸腫瘤干細胞的惡性轉化和腫瘤轉移的分子機制,并開展臨床轉化研究。

2)干細胞發育、衰老和融合的信號調控。實驗室的最新成果揭示了首個永久性的阻斷體內細胞融合的動物模型。在此基礎上,我們將進一步探討細胞融合與衰老的分子機理。

3)肌肉萎縮癥,糖尿病和肥胖的靶向治療。實驗室已發表的研究證實了新的疾病治療靶點。為此,我們將運用高通量篩選實驗找尋靶點基因的小分子抑制劑,進行臨床前期研究。

細胞通過感知和轉導機械力信號來產生恰當的胞內生物化學應答。細胞內外的機械力都有助于胞質分裂的成功,從而使一個細胞分裂為兩個子細胞。研究表明細胞胞質分裂的失敗與基因組不穩定性、腫瘤進化和衰老緊密關聯,而胞質分裂的成功則與細胞動力學和細胞外基質的相互作用密切相關。我們的研究目的是探究胞質分裂在衰老或其它病理狀況下的對細胞外基質不同剛度反應的機械力學和動力學機制。為此,我的實驗室聯合采用化學遺傳學、光遺傳學和高分辨率活細胞顯微成像技術探究胞質分裂失敗產生的結果和胞質分裂的機械生物學機制。

Cells sense and transduce mechanical signals to generate appropriate biochemical and cellular response. Both the cellular and extracellular mechanics contribute to the success of cytokinesis, which is the division of one cell into two. Failure of cytokinesis is implicated in genomic instability, tumor evolution, and aging. The success of cytokinesis is linked to the proper interaction of cytokinetic machinery and the extracellular matrix. We aim to study the mechanics and dynamics of cytokinesis in response to different extracellular matrix stiffness during aging or other pathological conditions. To this end, my lab couple chemical genetics, optogenetics and high resolution live cell microscopy imaging to study the mechanobiology of cytokinesis and the consequences of its failure.

The centrosome is an important signalling and microtubule organizing centre in mammalian cells. Centrosomal defects are a common in high grade cancer and have been shown to trigger tumorigenesis in mice. The impact of centrosomal defects in deregulating the cell cycle in cancer is poorly understood. In addition, cancer genome sequencing efforts worldwide show mutations on centrosomal components are common. Using genetic, cell biology and biochemical assays, we aim to study how the changes in centrosomal signalling lead to the deregulation of the cell cycle in cancer.

Salary is commensurate with qualifications and experience, compliant with policies for postdoctoral recruitment in Zhejiang University. Annual pay is in the region of 300,000 RMB.
